Submitter rationale: The variant satisfies PM2 criteria; Extremely low frequency in gnomAD population databases. However, the variant is present in heterozygous state in an individual that has squamous cell carcinoma of the tongue. But, due to lack of sufficient clinical evidence, the variant should be classified as a variant of uncertain significance.
