This variant lies in the FLNC gene (transcript NM_001458.5) at coding-DNA position 2072, where C is replaced by T; at the protein level this means replaces threonine at residue 691 with isoleucine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with FLNC-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces threonine with isoleucine at codon 691 of the FLNC protein (p.Thr691Ile). The threonine residue is moderately conserved and there is a moderate physicochemical difference between threonine and isoleucine.
