NM_000492.4(CFTR):c.1909C>A (p.Gln637Lys) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q637K variant (also known as c.1909C>A), located in coding exon 14 of the CFTR gene, results from a C to A substitution at nucleotide position 1909. The glutamine at codon 637 is replaced by lysine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
