Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005228.5(EGFR):c.3250G>A (p.Asp1084Asn), citing Ambry Variant Classification Scheme 2023: The p.D1084N variant (also known as c.3250G>A), located in coding exon 27 of the EGFR gene, results from a G to A substitution at nucleotide position 3250. The aspartic acid at codon 1084 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
