This variant lies in the RAG1 gene (transcript NM_000448.3) at coding-DNA position 318, where C is replaced by A; at the protein level this means replaces asparagine at residue 106 with lysine — a missense variant. Submitter rationale: This sequence change replaces asparagine, which is neutral and polar, with lysine, which is basic and polar, at codon 106 of the RAG1 protein (p.Asn106Lys). This variant is present in population databases (rs367725290, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with RAG1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1052679).
