Uncertain significance for Neuronal ceroid lipofuscinosis 11; GRN-related frontotemporal lobar degeneration with Tdp43 inclusions — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002087.4(GRN):c.100C>T (p.Pro34Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with serine, which is neutral and polar, at codon 34 of the GRN protein (p.Pro34Ser). This variant is present in population databases (rs748147151, gnomAD 0.009%). This missense change has been observed in individual(s) with behavioral variant frontotemporal dementia (PMID: 24022032). ClinVar contains an entry for this variant (Variation ID: 1052582). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
