Uncertain significance for Familial cancer of breast; Fanconi anemia complementation group J — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_032043.3(BRIP1):c.2861A>G (p.Lys954Arg), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the BRIP1 gene (transcript NM_032043.3) at coding-DNA position 2861, where A is replaced by G; at the protein level this means replaces lysine at residue 954 with arginine — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This sequence change replaces lysine with arginine at codon 954 of the BRIP1 protein (p.Lys954Arg). The lysine residue is weakly conserved and there is a small physicochemical difference between lysine and arginine. This variant has not been reported in the literature in individuals with BRIP1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
