This variant lies in the CHD2 gene (transcript NM_001271.4) at coding-DNA position 2749, where A is replaced by G; at the protein level this means replaces threonine at residue 917 with alanine — a missense variant. Submitter rationale: This sequence change replaces threonine with alanine at codon 917 of the CHD2 protein (p.Thr917Ala). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and alanine. This variant is present in population databases (rs766507274, ExAC 0.01%). This variant has not been reported in the literature in individuals with CHD2-related conditions.
