Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_020975.6(RET):c.3112A>T (p.Thr1038Ser), citing Ambry Variant Classification Scheme 2023: The p.T1038S variant (also known as c.3112A>T), located in coding exon 19 of the RET gene, results from an A to T substitution at nucleotide position 3112. The threonine at codon 1038 is replaced by serine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
