Uncertain significance for Developmental and epileptic encephalopathy, 26 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004975.4(KCNB1):c.676_685del (p.Asn226fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change results in a premature translational stop signal in the KCNB1 gene (p.Asn226Trpfs*18). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 633 amino acids of the KCNB1 protein. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with KCNB1-related conditions. This variant is not present in population databases (ExAC no frequency).
