This variant lies in the GABRB3 gene (transcript NM_000814.6) at coding-DNA position 522, where C is replaced by G; at the protein level this means replaces asparagine at residue 174 with lysine — a missense variant. Submitter rationale: This sequence change replaces asparagine with lysine at codon 174 of the GABRB3 protein (p.Asn174Lys). The asparagine residue is highly conserved and there is a moderate physicochemical difference between asparagine and lysine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with GABRB3-related conditions.
