Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_005188.4(CBL):c.1168G>C (p.Asp390His), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the CBL gene (transcript NM_005188.4) at coding-DNA position 1168, where G is replaced by C; at the protein level this means replaces aspartic acid at residue 390 with histidine — a missense variant. Submitter rationale: Variant summary: CBL c.1168G>C (p.Asp390His) results in a non-conservative amino acid change located in the Zinc finger, RING-type domain (IPR001841) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251350 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1168G>C in individuals affected with Noonan Syndrome And Related Conditions and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
