NM_000890.5(KCNJ5):c.1248G>C (p.Arg416Ser) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R416S variant (also known as c.1248G>C), located in coding exon 2 of the KCNJ5 gene, results from a G to C substitution at nucleotide position 1248. The arginine at codon 416 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species, and serine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
