The variant was identified in the UMD 26X as a neutral variant, where it was listed in co-occurrence with three different pathogenic BRCA1 variants and one pathogenic BRCA2 variant, increasing the likelihood that the c.441+36_441+49del variant does not have clinical significance. The variant was not identified in any other databases searched, including HGMD, Clinvar, COSMIC, and LOVD. The BRCA1 c.441+36_441+49del variant occurs outside of the splicing consensus sequence and in silico or computational prediction software programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, GeneSplicer, HumanSpliceFinder) do not predict a difference in splicing. In summary, based on the above information, this variant meets our laboratory's criteria to be classified as benign
