This variant lies in the TTN gene (transcript NM_133379.5) at coding-DNA position 15056, where A is replaced by C; at the protein level this means replaces histidine at residue 5019 with proline — a missense variant. Submitter rationale: The TTN p.His5019Pro variant was not identified in the literature nor was it identified in dbSNP, ClinVar or in the following control databases: the 1000 Genomes Project, the NHLBI GO Exome Sequencing Project, the Exome Aggregation Consortium (August 8th 2016), or the Genome Aggregation Database (March 6, 2019, v2.1.1). The p.His5019 residue has limited species conservation information and computational analyses (PolyPhen-2, BLOSUM, MutationTaster) provide inconsistent predictions regarding the impact to the protein.
