This variant lies in the TCIRG1 gene (transcript NM_006019.4) at coding-DNA position 1793, where C is replaced by G; at the protein level this means replaces alanine at residue 598 with glycine — a missense variant. Submitter rationale: The TCIRG1 p.Ala300Gly variant was not identified in the literature nor was it identified in dbSNP, ClinVar, Cosmic or LOVD 3.0. The variant was also not identified in the following control databases: the 1000 Genomes Project, the NHLBI GO Exome Sequencing Project, the Exome Aggregation Consortium (August 8th 2016), or the Genome Aggregation Database (Feb 27, 2017). The variant occurs outside of the splicing consensus sequence and 3 of 4 in silico or computational prediction software programs (MaxEntScan, NNSPLICE, GeneSplicer) do not predict a greater than 10% difference in splicing.
