This variant lies in the CDH1 gene (transcript NM_004360.5) at coding-DNA position 2052, where C is replaced by A; at the protein level this means replaces serine at residue 684 with arginine — a missense variant. Submitter rationale: The CDH1 p.Ser684Arg variant was not identified in the literature nor was it identified in the dbSNP, ClinVar, Cosmic, MutDB, Insight Colon Cancer Gene Variant Database, Zhejiang Colon Cancer Database, databases. The variant was not identified in the following control databases: the 1000 Genomes Project, the NHLBI GO Exome Sequencing Project, the Exome Aggregation Consortium (August 8th 2016), or the Genome Aggregation Database (Feb 27, 2017). The p.Ser684 residue is not conserved in mammals and four out of five computational analyses (PolyPhen-2, SIFT, AlignGVGD, BLOSUM, MutationTaster) do not suggest a high likelihood of impact to the protein; however, this information is not predictive enough to rule out pathogenicity.
