The variant was identified in dbSNP (ID: rs771708962) and in control databases in 5 of 250872 chromosomes at a frequency of 0.00001993 (Genome Aggregation Database March 6, 2019, v2.1.1). The variant was observed in the South Asian population in 5 of 30614 chromosomes (freq: 0.000163), but was not observed in the African, Latino, Ashkenazi Jewish, East Asian, European (Finnish), European (non-Finnish), or Other populations. The p.Gly204 residue is conserved in mammals and computational analyses (PolyPhen-2, SIFT, AlignGVGD, BLOSUM, MutationTaster) provide inconsistent predictions regarding the impact to the protein; this information is not very predictive of pathogenicity. The variant occurs outside of the splicing consensus sequence and 2 of 4 in silico or computational prediction software programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, GeneSplicer) predict a greater than 10% difference in splicing; this is not very predictive of pathogenicity.
