Pathogenic for Malignant tumor of breast — the classification assigned by Department of Pathology and Laboratory Medicine, Sinai Health System to NM_007294.4(BRCA1):c.5407-73_5467+4del: The BRCA1 c.5333_5467del variant (chr:17 g.41199660_41201211del GRCh37) results in in-frame deletion of exons 22-23, although the precise breakpoints of this deletion were not determined, nor were the effects of this variant on the resulting mRNA or protein product determined. The BRCA1 c.5333_5467del variant was not identified in the literature nor was it identified in the dbSNP, ClinVar, LOVD 3.0, UMD-LSDB, the Exome Aggregation Consortium (August 8th 2016), or the Genome Aggregation Database (Feb 27, 2017). The predicted protein consequence of this variant is p.Asp1778_His1822del and we cannot rule out the possibility this variant produces a truncated functional protein product; however, this is the type of variant expected to cause the disorder. In summary, based on the above information this variant is classified as pathogenic.
