The variant was identified in dbSNP (ID: rs530083542) and in control databases in 3 of 250740 chromosomes at a frequency of 0.00001196 (Genome Aggregation Database March 6, 2019, v2.1.1). The variant was observed in the following populations: Other in 1 of 6116 chromosomes (freq: 0.000164) and South Asian in 2 of 30614 chromosomes (freq: 0.000065), but was not observed in the African, Latino, Ashkenazi Jewish, East Asian, European (Finnish) or European (non-Finnish) populations. The p.Met6 residue is not conserved in mammals and computational analyses (PolyPhen-2, SIFT, AlignGVGD, BLOSUM, MutationTaster) provide inconsistent predictions regarding the impact to the protein; this information is not very predictive of pathogenicity.
