Likely pathogenic for Mucopolysaccharidosis, MPS-IV-A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000512.5(GALNS):c.899-167A>G, citing Invitae Variant Classification Sherloc (09022015): This sequence change falls in intron 8 of the GALNS gene. It does not directly change the encoded amino acid sequence of the GALNS protein. This variant is not present in population databases (gnomAD no frequency). This variant has been observed in individual(s) with mucopolysaccharidosis type IVA (PMID: 30305043). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 1048484). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
