NM_000512.5(GALNS):c.423G>T (p.Trp141Cys) was classified as Likely pathogenic for Keratan sulfate excretion in urine; Coarse facial features; Mucopolysaccharidosis, MPS-IV-A by Foundation for Research in Genetics and Endocrinology, FRIGE's Institute of Human Genetics, citing ACMG Guidelines, 2015: A heterozygous missense variant in exon 5 of the GALNS gene that results in the amino acid substitution of Cystine for Tryptophan at codon 141 was detected. The observed variant c.423G>T has not been reported in the 1000 genomes and gnomAD databases. The in-silico prediction of the variant is disease causing by MutationTaster and DANN. In summary, the variant meets our criteria to be classified as likely pathogenic.
