NM_000350.3(ABCA4):c.3179A>C (p.Gln1060Pro) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: ABCA4 c.3179A>C (p.Gln1060Pro) results in a non-conservative amino acid change located in the ABC transporter-like, ATP-binding domain (IPR003439) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251178 control chromosomes. c.3179A>C has been reported in the literature in a compound heterozygous individual affected with Retinitis Pigmentosa (Maggi_2021). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 33546218). ClinVar contains an entry for this variant (Variation ID: 1048120). Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
