This variant lies in the COL7A1 gene (transcript NM_000094.4) at coding-DNA position 6341, where G is replaced by A; at the protein level this means replaces glycine at residue 2114 with aspartic acid — a missense variant. Submitter rationale: Variant summary: COL7A1 c.6341G>A (p.Gly2114Asp) results in a non-conservative amino acid change located in the Collagen triple helix repeat region (IPR008160) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251294 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.6341G>A has been reported in the literature in the presumed compound heterozygous state in at least 1 individual affected with Dystrophic Epidermolysis Bullosa, Recessive (example, Cuadrado-Corrales_2010). These data do not allow any conclusion about variant significance for COL7A1-related conditions. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
