Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. ClinVar contains an entry for this variant (Variation ID: 1047970). This variant is also known as 325insCG. This premature translational stop signal has been observed in individuals with dystrophic epidermolysis bullosa (PMID: 9326325, 22209565, 31001817, 35979658). This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Glu109Alafs*39) in the COL7A1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in COL7A1 are known to be pathogenic (PMID: 16971478).
