Submitter rationale: The p.R157H variant (also known as c.470G>A), located in coding exon 4 of the RINT1 gene, results from a G to A substitution at nucleotide position 470. The arginine at codon 157 is replaced by histidine, an amino acid with highly similar properties. This alteration was reported in 0/2024 breast and/or ovarian cancer cases and 1/1886 unaffected controls (Li N et al. Breast Cancer Res Treat, 2016 09;159:385-92). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
