Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000059.4(BRCA2):c.431T>C (p.Val144Ala), citing Ambry Variant Classification Scheme 2023: The p.V144A variant (also known as c.431T>C), located in coding exon 4 of the BRCA2 gene, results from a T to C substitution at nucleotide position 431. The valine at codon 144 is replaced by alanine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
