Uncertain significance for Hypohidrotic X-linked ectodermal dysplasia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001399.5(EDA):c.950T>G (p.Phe317Cys), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the EDA gene (transcript NM_001399.5) at coding-DNA position 950, where T is replaced by G; at the protein level this means replaces phenylalanine at residue 317 with cysteine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with EDA-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces phenylalanine with cysteine at codon 317 of the EDA protein (p.Phe317Cys). The phenylalanine residue is highly conserved and there is a large physicochemical difference between phenylalanine and cysteine.
