This variant lies in the TERT gene (transcript NM_198253.3) at coding-DNA position 2612, where T is replaced by G; at the protein level this means replaces leucine at residue 871 with tryptophan — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This sequence change replaces leucine with tryptophan at codon 871 of the TERT protein (p.Leu871Trp). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and tryptophan. This variant has not been reported in the literature in individuals with TERT-related conditions.
