Uncertain significance for Hereditary spastic paraplegia 11 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_025137.4(SPG11):c.3814A>G (p.Met1272Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine with valine at codon 1272 of the SPG11 protein (p.Met1272Val). The methionine residue is moderately conserved and there is a small physicochemical difference between methionine and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with SPG11-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
