Uncertain significance for Juvenile polyposis syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004329.3(BMPR1A):c.590A>C (p.Glu197Ala), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with BMPR1A-related conditions. This variant is present in population databases (rs763548019, ExAC 0.003%). This sequence change replaces glutamic acid with alanine at codon 197 of the BMPR1A protein (p.Glu197Ala). The glutamic acid residue is moderately conserved and there is a moderate physicochemical difference between glutamic acid and alanine. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
