NM_005228.5(EGFR):c.1292A>G (p.Lys431Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K431R variant (also known as c.1292A>G), located in coding exon 11 of the EGFR gene, results from an A to G substitution at nucleotide position 1292. The lysine at codon 431 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
