Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000222.3(KIT):c.1652C>T (p.Pro551Leu), citing Ambry Variant Classification Scheme 2023: The p.P551L variant (also known as c.1652C>T), located in coding exon 11 of the KIT gene, results from a C to T substitution at nucleotide position 1652. The proline at codon 551 is replaced by leucine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
