Uncertain significance for Congenital contractural arachnodactyly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001999.4(FBN2):c.6046G>C (p.Asp2016His), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the FBN2 gene (transcript NM_001999.4) at coding-DNA position 6046, where G is replaced by C; at the protein level this means replaces aspartic acid at residue 2016 with histidine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid with histidine at codon 2016 of the FBN2 protein (p.Asp2016His). The aspartic acid residue is highly conserved and there is a moderate physicochemical difference between aspartic acid and histidine. This variant also falls at the last nucleotide of exon 47 of the FBN2 coding sequence, which is part of the consensus splice site for this exon. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with FBN2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
