NM_001999.4(FBN2):c.3957C>G (p.Asp1319Glu) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1319E variant (also known as c.3957C>G), located in coding exon 30 of the FBN2 gene, results from a C to G substitution at nucleotide position 3957. The aspartic acid at codon 1319 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
