Uncertain significance for Breast-ovarian cancer, familial, susceptibility to, 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002878.4(RAD51D):c.392A>C (p.Asn131Thr), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the RAD51D gene (transcript NM_002878.4) at coding-DNA position 392, where A is replaced by C; at the protein level this means replaces asparagine at residue 131 with threonine — a missense variant. Submitter rationale: This sequence change replaces asparagine with threonine at codon 131 of the RAD51D protein (p.Asn131Thr). The asparagine residue is weakly conserved and there is a small physicochemical difference between asparagine and threonine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with RAD51D-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
