Uncertain significance for Catecholaminergic polymorphic ventricular tachycardia 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001035.3(RYR2):c.7025G>T (p.Gly2342Val), citing Invitae Variant Classification Sherloc (09022015): Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt RYR2 protein function. This variant has been observed in individual(s) with clinical features of catecholaminergic polymorphic ventricular tachycardia (Invitae). This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with valine at codon 2342 of the RYR2 protein (p.Gly2342Val). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and valine. This variant disrupts the p.Gly2342 amino acid residue in RYR2. Other variant(s) that disrupt this residue have been observed in individuals with RYR2-related conditions (PMID: 29434162), which suggests that this may be a clinically significant amino acid residue. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
