This variant has not been reported in the literature in individuals with ATP6V0A4-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This sequence change replaces alanine with glutamic acid at codon 752 of the ATP6V0A4 protein (p.Ala752Glu). The alanine residue is highly conserved and there is a moderate physicochemical difference between alanine and glutamic acid.
