Uncertain significance for Familial temporal lobe epilepsy 7; Norman-Roberts syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005045.4(RELN):c.3139A>C (p.Ile1047Leu), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with RELN-related conditions. This variant is present in population databases (rs778716913, gnomAD 0.003%). This sequence change replaces isoleucine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 1047 of the RELN protein (p.Ile1047Leu). ClinVar contains an entry for this variant (Variation ID: 1043160). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The leucine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
