Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.3328A>C (p.Ile1110Leu), citing Ambry Variant Classification Scheme 2023: The p.I1110L variant (also known as c.3328A>C), located in coding exon 20 of the DICER1 gene, results from an A to C substitution at nucleotide position 3328. The isoleucine at codon 1110 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
