This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 3959, where C is replaced by G; at the protein level this means replaces serine at residue 1320 with cysteine — a missense variant. Submitter rationale: This sequence change replaces serine with cysteine at codon 1320 of the BRCA1 protein (p.Ser1320Cys). The serine residue is weakly conserved and there is a moderate physicochemical difference between serine and cysteine. This variant is not present in population databases (ExAC no frequency). This variant has been reported in individuals in the Leiden Open-source Variation Database (PMID: 21520333).
