Uncertain significance for Epidermolysis bullosa simplex 3, localized or generalized intermediate, with BP230 deficiency; Hereditary sensory and autonomic neuropathy type 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001374736.1(DST):c.17595G>C (p.Arg5865=), citing Invitae Variant Classification Sherloc (09022015): The DST gene has multiple clinically relevant transcripts. This variant occurs in alternate transcript NM_015548.4, and corresponds to NM_001723.5:c.*86069G>C in the primary transcript. This sequence change affects codon 3242 of the DST mRNA. It is a 'silent' change, meaning that it does not change the encoded amino acid sequence of the DST protein. This variant also falls at the last nucleotide of exon 47, which is part of the consensus splice site for this exon. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with DST-related conditions. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
