Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003977.4(AIP):c.321C>G (p.Asn107Lys), citing Ambry Variant Classification Scheme 2023: The p.N107K variant (also known as c.321C>G), located in coding exon 3 of the AIP gene, results from a C to G substitution at nucleotide position 321. The asparagine at codon 107 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
