NM_000038.6(APC):c.5182A>G (p.Ile1728Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I1728V variant (also known as c.5182A>G), located in coding exon 15 of the APC gene, results from an A to G substitution at nucleotide position 5182. The isoleucine at codon 1728 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Based on the available evidence, the clinical significance of this variant remains unclear.
