Uncertain significance for Sulfite oxidase deficiency due to molybdenum cofactor deficiency type A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001358530.2(MOCS1):c.11G>A (p.Arg4Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 4 of the MOCS1 protein (p.Arg4Gln). This variant is present in population databases (rs760690355, gnomAD 0.1%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with MOCS1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1041646). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
