NM_006231.4(POLE):c.6625A>G (p.Lys2209Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K2209E variant (also known as c.6625A>G), located in coding exon 47 of the POLE gene, results from an A to G substitution at nucleotide position 6625. The lysine at codon 2209 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
