Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.1844G>T (p.Trp615Leu), citing Ambry Variant Classification Scheme 2023: The p.W615L variant (also known as c.1844G>T), located in coding exon 10 of the ALK gene, results from a G to T substitution at nucleotide position 1844. The tryptophan at codon 615 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
