Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015047.3(EMC1):c.1027-3_1027-2del, citing Invitae Variant Classification Sherloc (09022015): This sequence change falls in intron 9 of the EMC1 gene. It does not directly change the encoded amino acid sequence of the EMC1 protein. It affects a nucleotide within the consensus splice site. This variant is present in population databases (rs747574162, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with EMC1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1041162). Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Experimental studies and prediction algorithms are not available or were not evaluated, and the effect of this variant on mRNA splicing is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
