Uncertain significance for Cystinuria — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000341.4(SLC3A1):c.1157A>G (p.Tyr386Cys), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs764849767, ExAC 0.001%). This sequence change replaces tyrosine with cysteine at codon 386 of the SLC3A1 protein (p.Tyr386Cys). The tyrosine residue is weakly conserved and there is a large physicochemical difference between tyrosine and cysteine. This variant has not been reported in the literature in individuals with SLC3A1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
