Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_017636.4(TRPM4):c.1035G>T (p.Glu345Asp), citing Ambry Variant Classification Scheme 2023: The p.E345D variant (also known as c.1035G>T), located in coding exon 8 of the TRPM4 gene, results from a G to T substitution at nucleotide position 1035. The glutamic acid at codon 345 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
